Stable isotopes in carotenoid biochemistry

چکیده

The use of stable isotopes in studies of carotenoid biosynthesis is reviewed, beginning with early work with l8 which defined the origin of oxygen in a range of xanthophylls. A series of experiments from our own laboratory is then described, in which 2H and 13C have been used to investigate the stereochemistry of cyclization to form C40 and C50 and i-ring carotenoids. Novel procedures involving 2H labelling from 2H20 have been used to demonstrate carotenoid transformations during biosynthesis and chloroplast development and to define the stereochemistry of the H attack at C-2 which initiates carotenoid cyclization. Procedures by which microorganisms are allowed to metabolize ['HI-glucose, in 2H20, or [13C6]glucose to make endogenous specifically labelled substrates are described. These methods allow the carotenoid C-I methyl substituents to be distinguished even when incorporation of exogenous labelled substrates is not possible, and have been used to show that the stereochemistry of cyclization to form the p and C-rings of the C40 carotenoids zeaxanthin and lutein is the same.

Carotenoid and retinoid metabolism: insights from isotope studies.

Use of isotopes as tracers has had an important role in elucidating key features of vitamin A and retinoid metabolism in animal models and humans. Their use has shown that beta-carotene absorption is variable, and that the appearance of beta-carotene and its metabolites in the blood by time since dosing follows characteristic patterns. Retinol formed from beta-carotene shows a different pattern...

Stable Isotope Application in Animal Nutrition Science

The application of stable isotope analysis (SIA) has become a standard scientific approach in Agricultural and Ecological researches and, more in general, in several disciplines such as biology, botany, zoology, organic chemistry, climatology, and nutrition. The main objectives of this paper are (1) to provide a simple definition of stable isotopes and (2) to illustrate analytical measurement m...

Climatic and Environmental Reconstruction Based on Stable Isotopes of Parishan Lake (Iran)

The isotopic documents of lake deposition are suitable potential means for studyingthe intercontinental, environmental and climatic conditions. In order to study theenvironmental and climatic conditions of Parishan area in Fars province in Holocene, a Corewas taken with the length of 2m from the Parishan lake sediment. From this core, kernel timeseries was provided from documents of carbon and ...

The Effect of the Crocus Sativus L. Carotenoid, Crocin, on the Polymerization of Microtubules, in Vitro

Objective(s): Crocin, as the main carotenoid of saffron, has shown anti-tumor activity both in vitro and in vivo. Crocin might interact with cellular proteins and modulate their functions, but the exact target of this carotenoid and the other compounds of the saffron have not been discovered yet. Microtubular proteins, as one of the most important proteins inside the cells, have several functio...

Stable isotopes link diet to avian yolk carotenoid allocation: a comparative study of five auk species (Charadriiformes: Alcidae).

The allocation of important but environmentally limited nutrients, such as carotenoids, often represents a trade-off between homeostasis and reproduction. However, key questions remain about how diet and species traits influence carotenoid allocation. We studied yolk carotenoid profiles and yolk color in relation to trophic level (based on delta(15)N values) in five species of seabirds belongin...
